| 靶点详情: | 叶酸受体(FR)为糖基化磷脂酰肌醇(GPI)锚定的细胞膜糖蛋白,是介导细胞摄取叶酸及叶酸类拮抗剂的高亲和力转运蛋白,主要包含FRα(FOLR1)、FRβ、FRγ 等亚型,其中FRα(FOLR1) 在人体中表达调控最具特异性与疾病相关性。生理状态下,FRα 在正常组织中呈低表达或限制性表达,而在儿童发育异常、遗传性叶酸代谢障碍、儿童实体肿瘤、血液系统疾病中呈显著高表达,通过调控叶酸摄取、DNA 合成与修复、细胞增殖分化、甲基化代谢等关键通路,广泛参与细胞周期进程与组织发育调控。 FRα 在儿童相关疾病中具有高度特异性:叶酸代谢紊乱可直接导致胚胎神经管发育缺陷、儿童生长发育迟缓、造血功能异常及神经系统损伤;FRα 异常高表达则与儿童肿瘤细胞增殖、侵袭及化疗耐药密切相关。其表达水平的动态变化可直接反映儿童机体叶酸代谢状态、细胞损伤程度与疾病进展进程,是儿童叶酸代谢障碍、发育性疾病、儿童肿瘤等领域极具特异性的核心生物标志物,也是儿童疾病机制研究、靶向干预探索与药物筛选的关键靶点。 |
